El Museo exhibe una de las mejores y más completas muestras, en toda América, de la pintura barroca italiana y de la escuela inglesa — Flaming June, un óleo por Frederick Lord Leighton– incluyendo, además, una excelente colección de la academia francesa y de la pintura española del siglo de oro. Desde el año 1979 el Museo de Arte de Ponce cuenta con el primer y único Laboratorio de Conservación de Puerto Rico, destinado a conservar la colección de pinturas, esculturas y obras en papel, propiedad de la institución.
